A search of the 1850 census turns up several Mary Drennans, but only three are of an age appropriate to be a sister of Matilda: Mary Drennan 39 TN wife of E C Drennan 41 PA, living in Haywood Co TN; Mary Drennan 39 KY wife of Charles Drennan 38 TN, in Macoupin Co IL; and Mary Drennan 43 KY wife of Ezekiel Drennan 48 SC, in Sangamon Co IL.
A search for Polly Drennan turns up only one, age 40 born in IL, married to Samuel Drennan 31 IL, living in Sangamon Co IL.
(Note that Sangamon Co IL is where Mary's sister Malinda married.)

A search of RootsWeb for any Drennan marrying a Mary or Polly turns up hundreds of matches. Restricting the Drennan to the names or ages above narrows the results to:
E C: no matches
Charles: only one, married to Mary Pit(t)mann
Ezekiel: only one, married to Mary Viney
Samuel: only one, married to Mary A Baker

It looks to me like this Mary must be the one who married Samuel Drennan.
